This example illustrates using WorkOut 2.5 to process WIZARD measurements with a typical quantitative assay.
In the example RIA assay vials containing B0, NSB, Blank and Standard concentrations are measured (4 replicates). Unknown samples are measured in duplicate. The assay calculations subtract the mean of the Blank measurements from each sample and calculates each samples’ % Binding using the B0 and NSB measurements. The % Bindings value are plotted against the specified concentrations and used to calculate the concentrations of the unknowns.
A WorkOut 2.5 RIA protocol for this scenario has been produced and is available for download here. This protocol uses a 10x5 virtual plate representation.
Also included are two example raw data files. Simply launch the RIA.APR file and select a file to process. The example data file “000018.csv” contains measurements from 50 vials and is represented with 1 virtual plate. The example data file “000029.csv” contains more samples and is represented with 5 virtual plates.
