For applications requiring replicating the samples across different plates:
It is possible to setup the software to measure and analyse replicates that are read on different plates. For example, you may have 96 samples each read on 3 separate plates (e.g. Unknown1 in wells A1 of the 3 plates to read.) Use the multiple plate option to setup the microplates using the same layout.
It is important to note that the software automatically numbers samples on different plates with contiguous numbering, in this example the first plate's samples are reported as Unknown1-96 and plate 2 and Unknown97-192. However, the analysis can be setup to perform the replicate calculations on the samples on different plates.
To find the average of the wells from each of the 3 plates:
Create a Matrix Expression transform, under the Expression tab, enter:
With this transform, the expression is evaluated on every well of every plate.
“w.1.1” refers to the well on the 1st matrix (raw) of the 1st plate and "w.1.2" refers to the well on the 1st raw matrix of the second plate. So you can see that the expression finds the average of the wells on each of the 3 plates.
Similarly you can calculate the %CV across the 3 plates using:
It is important to understand that the software uses the idea of matrices for the transforms and EACH plate has the same number of matrices. In this situation data is being averaged from 3 plates so there will be a matrix for each plate – this means that the results will be repeated in each plate of the table.
