Use the Generic Wizard to get started.
Setup the Data Acquisition as required to take the measurements at the two required wavelengths. (Note, with some instrumentation a ratio or difference calculation may be performed automatically by the instrument if you need both matrices of raw data available ensure that the Wizard displays 2 raw data items Raw1-Raw2)
Continue through the Wizard setting up the microplate layout and other settings as required.
Run the protocol for the first time.
After the measurements have been completed press the Edit Transforms button.
If a %CV transform has been added, select it and press Remove.
Create a new transform.
Select the Dual Matrix Expression transform
Under the Input Matrices tab select the two matrices, Raw 1 corresponds to x and Raw 2 corresponds to y.
Under the Expressions tab you will see the default Expression is x-y, this performs the difference calculation between the first read and the second. Accept this for a difference calculation (or edit accordingly)
Press OK to finish editing the transform and press OK to return to the main screen.
The Recalculate button will flash to alert you that the changes have not calculated yet. Press Recalculate to calculate the results using the new settings.
