I want to run a Quantitative assay with the standards measured in a different location on each plate read.

Follow these instructions to create a Quantitative assay which uses a different microplate layout on each plate that is read. Here the standards occur on every read plate but in a different orientation.

The calculations will be performed on a plate by plate basis, i.e. a curve fit will be made for each plate and the concentrations will be calculated using a “local” curve fit.

Please note build 294 or later is required (contact Dazdaq Solutions if you require an update).

From the Organiser select Create a new protocol. Select the Quantitative Wizard. Setup the Data Acquisition as required. On the Microplate Layouts step, press the Advanced button and ensure that the Create Multiple Plate Microplate Layouts control is ticked. Returning to the Microplate Layouts step, press Create.

From the Microplate Layout Editor: press the Add Layout button to add a layout for each plate to be read. Next edit the first plate layout by double clicking on it and setting up as required. Press the Next > button to edit the next plate layout. Remember that the Standards can occur in any location on each plate; however the same number of standards groups must occur on each plate. When you have setup all of the layouts press Save and Close to exit the Microplate Layout editor and return to the Quantitative Wizard.

Continue to the next step in the Quantitative wizard. The Standard Set Selection step will display the option “Perform a fit on every plate using local standards”, if each microplate layout was setup with the same number of standards on every plate this will be ticked by default. Now, continue through the wizard entering the settings for your Quantitative assay and launch the protocol ready to run.

Each time you run the protocol you can press the Start button as normal to start the measurements.

After measurements are completed you can view the fit on each plate under then Analysis tab, you can also overlay the fits from each plate.
